Marijuana-Like Chemicals in the Brain
Calm Neurons



Stanford, Calif. -- From the munchies to the giggles to paranoia, smoking marijuana causes widespread changes in the brain.

Now researchers at Stanford University School of Medicine are a step closer to understanding how the drug's active ingredients -- tetrahydrocannabinol and related compounds, called cannabinoids -- may exert their effects.

David Prince, MD, the Edward F. and Irene Thiele Pimley Professor of Neurology and Neurological Sciences, and his colleagues found that a group of neurons that act as information gatekeepers in the brain's major information processing center, called the cerebral cortex, release cannabinoids that quiet their own activity. This form of self-inhibition is a novel way for neurons to regulate their own ability to send messages to their neighbors. Tetrahydrocannabinol from marijuana may work its brain-altering magic by binding to these same cells.

"Marijuana is a major drug of abuse with actions in the brain that aren't entirely known. Now we understand one piece of the puzzle," Prince said. The work of Prince and his colleagues John R. Huguenard, PhD, associate professor of neurology and neurological sciences, and Alberto Bacci, PhD, staff research associate, is published in the Sept. 16 issue of Nature.

The cells under scrutiny lie in the cerebral cortex. This region processes information from the eyes, ears, skin and other sense organs, regulates movement and performs complex functions such as those involved in thinking, learning and emotions. The cortex contains two major types of nerve cells: pyramidal neurons that excite both local and more distant neighbors, and inhibitory interneurons that act as local dimming switches, shutting down the activity of nearby brain cells. The inhibitory interneurons prevent the brain from taking in and responding to every thought, sight or sound it encounters. They also protect against runaway excitation such as that seen in epilepsy.

In previous work, other researchers had found that pyramidal cells manufacture and release cannabinoids that bind to a receptor on the membrane of interneurons. In this process, called retrograde signaling, the pyramidal cell does the equivalent of slipping its guardian interneuron some sleeping pills. It frees itself from inhibition by releasing cannabinoids that briefly decrease the interneuron's ability to release inhibitory molecules.

In contrast, Bacci and his colleagues found that interneurons can drug themselves when they get repetitively excited, triggering a self-inhibition process. The class of interneurons the researchers studied, the so-called "LTS cells" of the cerebral cortex, manufacture and release cannabinoids that bind to their own cannabinoid receptors and shut down their ability to signal other neurons. By shutting themselves off, the interneurons block their quieting action on the excitatory pyramidal cells -- an effect that can last as long as 35 minutes, much longer than what had been seen with retrograde inhibition. Without the quieting effect, pyramidal cells signal more intensely, triggering a higher level of activity in circuits of the cortex.

Prince said it's too early to know exactly how marijuana binding to the cannabinoid receptor exerts its behavioral effects. However, because the interneurons inhibit cells that have such wide-ranging effects, it's no surprise that the drug alters how people perceive the world around them. "A loss of inhibition in pyramidal cells could produce changes in perception, in motor function and in everything the cerebral cortex does," he said.

The Stanford team hopes that by studying how these receptors work, researchers may learn how to make drugs that selectively bind and block subtypes of cannabinoid receptors on one type of cell but not another. This may be one way to harness the medically useful aspect of marijuana without causing brain-altering side effects.

According to Prince, such drugs could also be useful in treating epilepsy. Pyramidal cells are among those that fire out of control during a seizure. One reason these cells fire so rapidly may be that interneurons get shut down. A drug that blocks cannabinoid receptors on some types of inhibitory interneurons might allow them to continue quieting the pyramidal cells during periods of intense activity.

Why do they need to make a fake version when the real thing seems to work great? To change the brain altering side effects? What a load of crap! the brain altering side effects are what make it work.

It is a condition in the brain where your brain doesn't naturally produce enough cannabinoids and the Lord has provided a way for us to heal ourselves. There is a cure for anything that ails you in Nature.

Half of our problem in society today is that we are constantly poisoning ourselves with pharmaceuticals.

Why are viruses so strong today? Maybe because the doctor's "deal" penicillin every time somebody has a runny nose? I saw that the average cost per family in Canada for health care is 16,000 dollars per year. It makes me sick just thinking about it. They tell us all to drink milk, its good for us! They pump the dairy cows up with female hormones to produce more milk so they can make more money. I wonder why I had to but my nine year old daughter a bra? If I gave a shit if I was being politically correct I'd ask why is male homosexuality become so widespread? It couldn't be that they are poisoning us with their prescription poisons, or could it? I worked in a couple of big factory style meat production factories and we used to to put amoxycylin in the water to prevent them from getting sick. Is it so far fetched that all these virus's and bacteria could be developing an immunity to the drugs because of this kind of overuse? I don't think so! What about all the piils that don't get used or the prescriptions you don't finish that get flushed down toilets or washed down the sink? Where do they go? what effct does that have?

I'd say before they talk about how much testing God's medicine has been subjected to, they better look to themselves. I keep thinking to myself, weed heals everything. Every day I see my reality taking effect. I just read in these forums today that weed even cures freakin herpes or at least works in some benificial way. Epilepsy, multiple sclerosis, Cancer, arthritis, chronic pain, anorexia, all kinds of "diseases" affecting the nervous system. If your brain can't process all this information at once try a little ak47, it really help restrict those medical things they say prevent your brain from processing too much information. Excuse the outburst but i smoked a little cannabis concentrate tonite so my brain is wide open and I'm turning loose o you for a minute. Here's a good one for you. Is it in the best intersest on the rich and powerful, and their minions the politicians to have a populace that can process more information, be cognicant of their doublespeak, or to keep the masses, especially the slave class dumbed down with a depressant like alcohol? You may think I am some whacko consiracy theorist who is a little paranoid from his medicine, but ask yourself this. Is it paranoia if its the truth? If I have to play trust and I have to pick between man and Nature, I'll pick nature every time. God has provided for us all we need, and all these scientists would do well to remember that the poison apple that eve took was from the tree of knowledge. They think they have the knowledge to mess with nature but the will eventually be the end of mankind and life on earth as we know it.

We know how good is this herb DrGreenthumb :D , but our government is playing a head game with everyone by disinforming the public instead of telling the truth.

Why?

Well, it's a big money thing with pharmaceutical companies and they don't want too many people using cannabis and it's different strains...it's too much of a cheap medication and easy to grow.

Cannabis isn't as addictive and it's non-toxic. Cannabis does not damage internal organs like pharmaceuticals either.

The proper strain(s) of cannabis will also be a far superior analgesic compared to heroin, morphine, demerol, fentanyl, oxycontin, oxycodone, percocet, percodans and other very dangerous, addictive narcotics.

Over 400 cannabinoids are present in cannabis and each strain has different levels of these cannabinoids.

It's a big race for an incredible new generation of cannabis-based pharmaceutical market we will see in the years to come.

New companies will surge out of the market and old ones will die.

Many companies will change their views about cannabinoid properties and they will start to develop quite a range of cannabis-based medicines.

I suspect that there's a cure for cancer and many more diseases in cannabis.

It's a miracle plant and what stalled the progress in developping new non-toxic medications is this prohibition that has been lasting for already too long.

But we cannabis users are one or many steps ahead...aren't we?
